Determination of Astaxanthin from Hainan Penacus Orientalis with Supercritical CO2-reversed phase HPLC

Abstract
In the report, supercritical CO2 fluid extraction (SFE) was used to extract Astaxanthin from Hainan Penacus orientalis and four factors and three level orthogonal experimental design was used to optimize the SFE conditions. The results indicated that the optimum extraction pressure was 35 MPa, the optimum extraction temperature was 60℃, the optimum extraction time was 2 h. Reversed phase HPLC was used to determine Astaxanthin, the chromatographic column was waters novapak C18 column (3.9×150 mm, 5 um); methol-dichloromethane-water (50: 2: 3, V: V: V) was used as the mobile phase at a flow rate of 0.6 mL∙min-1; the detection was performed by a UV detector at 478 nm; the linear range of the method was 10-100 μg∙mL-1 (R2=0.999 2); the recovery rate was 99.01%.
