THE USE OF HPLC IN THE ANALYSTS OF AMINO ACID

  • Seventeen aminoa cids were determined qualitattively and quantitatively on Gilson C18 reverse phase column, based on monitoring of fluorescent derivatives of amino acids. Amino acid composition of ∝chain and β chain of human hemoglobine was analysed and quantitatively determined on the same column. Recoiery test of seventeen amino acids was performed and the recovery rate is 96.1—99.2%.
    Qualitativeanalysis showed that the elution peaks of most amino acids were seperated. The retention time of amino acids on the column was measured and its’ coe fficient of variation is 2.37%. The relation between the fluorescent density and the concentration of amino acids is discussed. The relative coefficient is 0.985.
    We conclude that HPLC can be used for quantitative analysis of amino acids instead of amino acid analyzer.
